Samantha Ronson was born into a wealthy family in London. Her mother is writer/socialite Ann Dexter-Jones and her father is real estate mogul Laurence Ronson. Her older brother is renowned music producer Mark Ronson, and her twin sister is fashion designer Charlotte Ronson. When her mother married Foreigner guitarist Mick Jones, her childhood became surrounded by music, thus her interest in the field. She was in a rap band called Low Lifes at the age of 16.

She had performed as a disc jockey for MTV's New Year's Eve 2001 show in Times Square. She also became the first rock act signed to Roc-A-Fella Records. She released four songs under the label Super, which garnered very little public interest. One of the songs under her first album is "Built This Way," which was featured in the 2004 film "Mean Girls."

She wrote and recorded her first album, "Red." It was never officially released, though the songs were available for download from her MySpace page.

Ronson became involved with actress Lindsay Lohan in 2008. Although they never implicitly stated that they are romantically involved together, they have always dropped hints, such as being affectionate in public, which the media interpreted as such.

She had been involved in a legal feud with celebrity blogger Perez Hilton after he blamed her for the drugs found in Lohan's vehicle in 2007. She lost the case on the grounds that Hilton was entitled on his 1st Amendment rights and that he did not act with malice. The court ordered her to pay Hilton $85,000 in legal fees.
